Efek Ekstrak Etanol Biji Ketumbar terhadap Oksidasi Protein Plasma Darah Tikus Obesitas akibat Diinduksi Diet Tinggi Lemak = Effect of Ethanol Extract of Coriander Seeds on Protein Oxidation in Blood Plasma ofObese Rats Induced by High-Fat Diet.

Latar belakang: Obesitas merupakan salah satu faktor risiko utama penyakit metabolik yang berhubungan erat dengan peningkatan stres oksidatif. Salah satu biomarker spesifik stres oksidatif adalah AOPP yang terbentuk akibat oksidasi protein plasma. Ekstrak biji ketumbar (Coriandrum sativum L.) diketahui mengandung flavonoid dan polifenol yang berperan sebagai antioksidan alami. Penelitian ini bertujuan untuk menilai efek pemberian ekstrak etanol biji ketumbar terhadap kadar AOPP pada tikus obesitas yang diinduksi diet tinggi lemak. Metode: Penelitian eksperimental in vivo ini menggunakan 25 ekor tikus Wistar jantan yang dibagi menjadi lima kelompok: kontrol normal, normal+ekstrak, obes + ekstrak selama induksi, obes, dan obes + ekstrak setelah induksi. Induksi obesitas dilakukan dengan diet tinggi lemak selama 24 minggu. Ekstrak biji ketumbar diberikan dengan dosis 100 mg/kgBB per hari. Kadar AOPP plasma darah diukur menggunakan spektrofotometri pada panjang gelombang 340 nm. Analisis statistik menggunakan One-Way ANOVA dilanjutkan dengan uji Tukey, dengan p < 0,05 dianggap bermakna. Hasil: Rata-rata kadar AOPP tertinggi terdapat pada kelompok obes (506,80 ± 104,02 μmol/L), sedangkan terendah pada kelompok kontrol normal (292,53 ± 80,66 μmol/L). Kelompok obes + ekstrak selama induksi menunjukkan penurunan bermakna kadar AOPP (329,20 ± 90,01 μmol/L) dibandingkan kelompok obes (p < 0,05). Kelompok obes + ekstrak setelah induksi (408,03 ± 115,23 μmol/L) juga menunjukkan kadar AOPP lebih rendah dibandingkan obes, namun tidak signifikan. Simpulan: Ekstrak biji ketumbar terbukti menurunkan kadar AOPP plasma darah tikus obesitas, terutama bila diberikan selama fase induksi obesitas. Hal ini mendukung potensi ketumbar sebagai antioksidan alami dalam pencegahan kerusakan protein akibat stres oksidatif pada obesitas.

Background: Obesity is a major risk factor for metabolic diseases and is strongly associated with increased oxidative stress. One of the specific biomarkers of oxidative stress is AOPP, formed through plasma protein oxidation. Coriander seeds (Coriandrum sativum L.) contain flavonoids and polyphenols with natural antioxidant properties. This study aimed to evaluate the effect of ethanol extract of coriander seeds on plasma AOPP levels in obese rats induced by a high-fat diet. Methods: An in vivo experimental study was conducted on 25 male Wistar rats, divided into five groups: normal control, normal + extract, obese + extract during induction, obese, and obese + extract after induction. Obesity was induced by a high-fat diet for 24 weeks. Coriander seed extract was administered at a dose of 100 mg/kgBW per day. Plasma AOPP levels were measured using spectrophotometry at a wavelength of 340 nm. Statistical analysis was performed using One-way ANOVA followed by Tukey’s test, with p < 0.05 considered significant. Results: The highest mean AOPP level was observed in the obese group (506.80 ± 104.02 μmol/L), while the lowest was in the normal control group (292.53 ± 80.66 μmol/L). The obese + extract during induction group showed a significant reduction in AOPP (329.20 ± 90.01 μmol/L) compared to the obese group (p < 0.05). The obese + extract after induction group (408.03 ± 115.23 μmol/L) also showed lower AOPP levels compared to the obese group, but the differences were not statistically significant. Conclusion: Ethanol extract of coriander seeds reduced plasma AOPP levels in obese rats, with the most significant effect observed in the obese + extract during induction group. These findings support the potential role of coriander seeds as a natural antioxidant to prevent protein oxidation associated with obesity.
