Analysis of Antioxidant Levels based on Degree of Malignancy in Patients with Type 1 Endometrial Cancer in RSCM

Introduction Endometrial cancer is frequently associated with oxidative stress, a condition marked by the imbalance between the generation of reactive metabolite and free radicals and the removal of these by antioxidants. Catalase, a heme oxidoreductase enzyme, plays as one of the body’s first line of defense against oxidative stress. This suggests a relationship between the specific activity of catalase enzyme and the degree of malignancy of endometrial cancer. Method An analytical cross sectional study using previously collected samples of type 1 endometrial cancer from patients in RSCM, which are categorized into grade 1, grade 2, and grade 3. First, the tissue homogenate is produced by centrifugation, and the supernatant is stored. Measurements of the specific activity of catalase were conducted via spectrophotometry at wavelengths 210nm. The total protein content of the sample is measured using The Warburg-Christian method via spectrophotometry at 210nm. Results No significant difference was observed between the specific activity of catalase between the degrees of malignancy (grade 1, grade 2, grade 3) in endometrial cancer (p > 0.05), with p = 0.2224, of patients in RSCM. However, the activity is shown to be highest in grade 3 endometrial cancer. Conclusion There were no differences in catalase levels according to the degree of malignancy in type 1 endometrial cancer tissue in patients at RSCM.

Latar Belakang Kanker endometrium sering dikaitkan dengan stres oksidatif, suatu kondisi yang ditandai dengan ketidakseimbangan antara produksi metabolit reaktif dan radikal bebas serta pembuangannya oleh antioksidan. Katalase, enzim heme oksidoreduktase, berperan sebagai salah satu garis pertahanan pertama tubuh untuk melawan stres oksidatif. Hal ini menunjukkan adanya hubungan antara aktivitas spesifik enzim katalase dengan derajat keganasan kanker endometrium. Metode Sebuah studi analitik cross-sectional menggunakan sampel kanker endometrium tipe 1 yang dikumpulkan sebelumnya dari pasien di RSCM, yang dikategorikan menjadi grade 1, grade 2, dan grade 3. Pertama, homogenat jaringan diproduksi dengan sentrifugasi, dan supernatan disimpan. Pengukuran aktivitas spesifik katalase dilakukan melalui spektrofotometri pada panjang gelombang 210 nm. Kandungan protein total sampel diukur menggunakan metode Warburg-Christian melalui spektrofotometri pada 210nm. Hasil Tidak terdapat perbedaan signifikan antara aktivitas spesifik katalase antara derajat keganasan (grade 1, grade 2, grade 3) pada kanker endometrium (p > 0,05) pasien di RSCM. Namun, aktivitas tersebut terbukti paling tinggi pada kanker endometrium tingkat 3. Kesimpulan Tidak terdapat perbedaan kadar katalase menurut derajat keganasan pada jaringan kanker endometrium tipe 1 pasien di RSCM.
