Analisis Efek Doksisiklin Terhadap Aktivitas Superoxide Dismutase Jantung Tikus Yang Mengalami Cardiac Remodelling = Analysis of The Effects of Doxycycline on Superoxide Dismutase (SOD) Activity in The Heart of Mice with Cardiac Remodelling.

Latar Belakang Cardiac remodelling adalah maladaptasi jantung yang berisiko tinggi gagal jantung, dipicu oleh hipertensi kronik dan tingginya stres oksidatif. Enzim antioksidan utama, Superoxide Dismutase (SOD), berperan penting dalam menurunkan stres oksidatif. Doksisiklin, antibiotik golongan tetrasiklin, memiliki sifat antioksidan. Penelitian ini bertujuan menganalisis efek doksisiklin terhadap aktivitas SOD pada jantung tikus yang mengalami cardiac remodelling Metode Penelitian in vivo ini menggunakan homogenat jaringan jantung tikus Sprague Dawley jantan. Model cardiac remodelling diinduksi melalui uninefrektomi, DOCA 20 mg/kgBB, dan NaCl 1% selama lima minggu. Terdapat empat kelompok perlakuan (n=5 per kelompok): Sham, DOCA, DOCA + DOXY 15 mg/kgBB, dan DOCA + DOXY 30 mg/kgBB. Aktivitas SOD diukur melalui spektrofotometri menggunakan EZ-SOD Assay Kit. Data yang terdistribusi normal dianalisis menggunakan uji One-Way ANOVA Hasil Kelompok DOCA menunjukkan penurunan rerata aktivitas SOD (91,51% ± 3,09246 SEM) dibandingkan kelompok Sham (98,05% ± 1,773202 SEM). Pemberian doksisiklin (dosis 15 mg/kgBB: 93,46%; dosis 30 mg/kgBB: 94,88%) menunjukkan tren peningkatan rerata aktivitas SOD dibandingkan kelompok DOCA. Namun, hasil uji One-Way ANOVA (p-value = 0,2901) menunjukkan bahwa tidak ada perbedaan aktivitas SOD yang bermakna secara statistik di antara keempat kelompok perlakuan Kesimpulan Aktivitas enzim Superoxide Dismutase (SOD) mengalami penurunan pada jantung tikus yang mengalami cardiac remodelling akibat uninefrektomi, pemberian DOCA, dan garam fisiologis. Doksisiklin dapat meningkatkan aktivitas enzim SOD pada jantung tikus yang diberi perlakuan tersebut, meskipun peningkatan ini tidak signifikan secara statistik, namun terdapat tren peningkatan aktivitas SOD pada kelompok yang diberikan doksisiklin

Introduction Cardiac remodelling is a maladaptive response, significantly increasing the risk of heart failure, primarily triggered by chronic hypertension and high oxidative stress. Superoxide Dismutase (SOD) is a key antioxidant enzyme crucial for reducing oxidative stres. Doxycycline, a tetracycline antibiotic, is known for its antioxidant and anti-inflammatory properties. This study aimed to analyze the effect of doxycycline administration on cardiac SOD activity in rats with cardiac remodelling. Method This in vivo study used stored heart tissue homogenates from male Sprague Dawley rats. The cardiac remodelling model was induced via uninephrectomy, DOCA 20 mg/kgBW, and 1% NaCl solution for five weeks. Four treatment groups were analyzed (n=5/group): Sham, DOCA, DOCA + DOXY 15 mg/kgBW, and DOCA + DOXY 30 mg/kgBW. SOD activity was measured spectrophotometrically. Normally distributed data were analyzed using One-Way ANOVA. Results The DOCA group (91.51% ± 3.09 SEM) showed reduced mean SOD activity compared to the Sham group (98.05% ± 1.77 SEM), suggesting elevated oxidative stress. Doxycycline (15 mg/kgBW: 93.46%; 30 mg/kgBW: 94.88%) demonstrated a trend of increased mean SOD activity relative to the DOCA group. However, the One-Way ANOVA result (p=0.2901) indicated no statistically significant difference in SOD activity among the groups. Conclusion Superoxide Dismutase (SOD) enzyme activity decreased in the hearts of rats that experienced cardiac remodelling due to uninephrectomy, DOCA administration, and physiological salt. Doxycycline has the potential to increase SOD enzyme activity in the cardiac remodelling rat model or can increase the SOD enzyme activity, although this increase was not statistically significant, but there was a trend of increased SOD activity in the group given doxycycline
