Analisis Perbandingan Ultrastruktur Sperma Setelah Kriopreservasi Dengan Krioprotektan Modifikasi dan Kitazato Menggunakan Scanning Electron Microscope = Comparative Analysis of Sperm Ultrastructure After Cryopreservation Using Modified and Kitazato Cryoprotectants via Scanning Electron Microscope.

Latar Belakang Kriopreservasi sperma sangat penting dalam teknologi reproduksi berbantu (TRB), namun proses freeze-thaw dapat menyebabkan kerusakan sperma. Saat ini krioprotektan yang tersedia di pasaran memiliki biaya yang mahal dengan ketersediaannya yang terbatas. Penelitian ini bertujuan membandingkan efektivitas modifikasi krioprotektan yang lebih terjangkau namun cukup efektif dalam mempertahankan kualitas sperma, yakni ultrastruktur sperma. Metode Penelitian eksperimental laboratorium ini menggunakan sembilan sampel sperma manusia normozoospermia, dibagi menjadi tiga kelompok, yaitu sperma segar (kontrol), kriopreservasi dengan Kitazato (komersial), dan kriopreservasi dengan krioprotektan modifikasi. Ultrastruktur sperma (kepala, midpiece, ekor) diamati dengan menggunakan scanning electron microscope (SEM) dan dilakukan penilaian untuk kelainannya. Data dianalisis dengan uji Kruskal-Wallis dan Mann-Whitney U. Hasil Rata-rata skor abnormalitas total untuk kelompok sperma segar, Kitazato, dan modifikasi masing-masing adalah 3,00; 2,67; dan 3,00. Analisis statistik menunjukkan tidak ada perbedaan signifikan pada skor total (p=0,786) maupun skor parameter individual (kepala, midpiece, ekor) antara ketiga kelompok. Perbandingan langsung antara kedua krioprotektan juga tidak menunjukkan perbedaan signifikan (p > 0,05). Kesimpulan Hasil penelitian menunjukkan tidak terdapat perbedaan bermakna pada tingkat abnormalitas ultrastruktur sperma antara krioprotektan Kitazato dan krioprotektan modifikasi. Dengan demikian, krioprotektan modifikasi memiliki efektivitas yang setara dengan Kitazato dalam mempertahankan integritas ultrastruktur sperma setelah proses kriopreservasi.

Introduction Sperm cryopreservation is essential for assisted reproductive technology (ART), yet the freeze-thaw process can induce sperm damage. Commercially available cryoprotectants, such as Kitazato, are often costly and have limited availability. This study aimed to compare the effectiveness of a more affordable modified cryoprotectant in preserving sperm quality, specifically sperm ultrastructure. Method This laboratory experimental study used nine normozoospermic human sperm samples, divided into three groups: fresh sperm (control), cryopreservation with commercial Kitazato, and cryopreservation with a modified cryoprotectant. Sperm ultrastructure (head, midpiece, tail) was observed using scanning electron microscope (SEM) and scored for abnormalities. Data were analyzed using the Kruskal-Wallis and MannWhitney U tests. Results The mean total abnormality scores for the fresh sperm, Kitazato, and modified cryoprotectant groups were 3,00; 2,67; and 3,00, respectively. Statistical analysis revealed no significant differences in the total score (p=0.786) or individual parameter scores (head, midpiece, tail) among the three groups. A direct comparison between the two cryoprotectants also showed no significant difference (p > 0.05). Conclusion The results showed no significant difference in the level of sperm ultrastructural abnormalities between the Kitazato and modified cryoprotectants. Therefore, the modified cryoprotectant demonstrated comparable effectiveness to Kitazato in preserving sperm ultrastructural integrity following cryopreservation.
