Analisis Kadar Mda Dan Gsh Organ Limpa Tikus Model Hemosiderosis Setelah Pemberian Kombinasi Deferiprone Dan Ekstrak Etanol Buah Mahkota Dewa = Analysis of MDA and GSH Levels in the Spleen ofHemosiderosis Rats Models after Administration of a Combination of Deferiprone and Ethanol Extract of Phaleria macrocarpa Fruit.

Latar Belakang. Besi berlebih akibat transfusi berulang menginduksi hemosiderosis. Besi tidak terikat transferrin (NTBI) mempromosikan reaksi Fenton yang menghasilkan ROS, memicu peroksidasi lemak di limpa sehingga MDA terbentuk. Tingginya kadar MDA merusak limpa mengganggu fungsi imunitas. Tubuh menghasilkan GSH, yaitu antioksidan untuk melawan kerusakan oksidatif. Deferiprone merupakan kelator besi standar yang penggunaannya dinilai banyak efek samping. Senyawa mangiferin pada buah mahkota dewa memiliki manfaat sebagai antioksidan dan kelator besi. Penelitian bertujuan menganalisis hubungan terapi kombinasi deferiprone dengan ekstrak etanol buah mahkota dewa terhadap kadar MDA serta GSH organ limpa tikus model hemosiderosis. Metode. Dua puluh ekor tikus Sprague-Dawley dibagi ke dalam 5 kelompok: normal (N), kontrol negatif (KN), deferiprone (D), Phaleria macrocarpa (PM), kombinasi deferiprone dosis penuh dan Phaleria macrocarpa (DPM-1), kombinasi deferiprone dosis setengah dan Phaleria macrocarpa (DPM-2). Semua kelompok kecuali normal mendapatkan injeksi besi intraperitoneal 15mg dengan frekuensi 2x/minggu selama 8 minggu. Terapi diberikan mulai minggu ke-4, limpa diambil pada minggu ke-8 dan dibuat homogenat. Kadar MDA diukur menggunakan spektrofotometri dan GSH diukur menggunakan metode kolorimetri. Hasil. Kadar MDA menurun pada semua kelompok terapi. Uji analisis statistik menunjukkan distribusi data normal (p > 0,05). Namun, varians tidak homogen dan tidak ada perbedaan signifikan antar kelompok sampel (p > 0,05). Terdapat tren peningkatan kadar GSH di semua kelompok setelah pemberian terapi kecuali DPM-2. Kelompok DPM-1 (625,39 μmol/L) mengalami peningkatan GSH terbanyak. Uji analisis statistik menunjukkan distribusi data tidak normal, varians tidak homogen dan perbedaan antar kelompok tidak signifikan. Kesimpulan. Pemberian kombinasi deferiprone dan Phaleria macrocarpa tidak menurunkan kadar MDA dan tidak meningkatkan GSH pada organ limpa tikus hemosiderosis.

Introduction Iron overload from repeated transfusions induces hemosiderosis. Non-transferrin-bound iron (NTBI) promotes the Fenton reaction, generating reactive oxygen species (ROS) that trigger lipid peroxidation in the spleen, thereby increasing MDA levels. Elevated MDA damages the spleen and impairs immune function. Glutathione (GSH) serves as an antioxidant defense. Deferiprone is a standard iron chelator and known for having side effects. Mangiferin, a bioactive compound in Phaleria macrocarpa, possesses both antioxidant and iron-chelating properties. This study analyzed the effects of combined therapy with deferiprone and ethanol extract of Phaleria macrocarpa towards MDA and GSH levels in the spleen of hemosiderosis rat models. Method Twenty Sprague-Dawley rats were divided into five groups: normal (N), negative control (KN), deferiprone (D), Phaleria macrocarpa (PM), combination of full-dose deferiprone and PM (DPM-1), and combination of half-dose deferiprone and PM (DPM-2). Except for the normal group, all rats received intraperitoneal iron injections (15 mg) twice weekly for eight weeks. Treatments were administered from fourth week, and spleen homogenates were collected at eighth week. MDA levels were measured using spectrophotometry, and GSH levels were assessed by colorimetric methods. Results MDA levels decreased in all treatment groups. Data were normally distributed (p > 0.05), showed non-homogeneous variance, with no significant intergroup differences (p > 0.05). There was a trend of increased GSH levels in all groups after treatment, except for DPM- 2. The DPM-1 group (625.39 μmol/L) showed the greatest increase in GSH levels. Statistical analysis indicated that the data distribution was not normal, the variance was not homogeneous, and the differences between groups were not significant. Conclusion Administration of combined deferiprone and Phaleria macrocarpa did not reduce MDA and did not increase GSH levels in the spleen of hemosiderosis-induced rats.
