Evaluasi Aktivitas Litik Isolat Bakteriofaga terhadap Staphylococcus aureus terkait Infeksi Sendi Periprostetik: Studi In Vitro = Evaluation of the Lytic Activity of Bacteriophage Isolates against Staphylococcus aureus Related to Periprosthetic Joint Infection: An In Vitro Study.

Pendahuluan: Infeksi sendi periprostetik (Periprosthetic Joint Infection, PJI) merupakan komplikasi serius pasca artroplasti sendi panggul dan lutut, dengan Staphylococcus aureus sebagai salah satu patogen tersering. Keberhasilan terapi PJI sering terhambat oleh pembentukan biofilm dan meningkatnya resistensi antibiotik, sehingga diperlukan pendekatan biologis alternatif, salah satunya terapi bakteriofag. Metode: Penelitian ini merupakan studi eksperimental in vitro. Sampel limbah Instalasi Pengolahan Air Limbah (IPAL) Rumah Sakit Cipto Mangunkusumo digunakan sebagai sumber isolasi bakteriofag. Aktivitas litik bakteriofag terhadap Staphylococcus aureus diuji menggunakan metode double agar overlay (plaque assay) dengan mengamati terbentuknya zona lisis (plaque) serta variasi morfologi plaque. Hasil: Isolat bakteriofag berhasil diperoleh dari sampel limbah IPAL dan menunjukkan aktivitas litik terhadap Staphylococcus aureus. Aktivitas tersebut ditandai dengan terbentuknya plaque dengan variasi ukuran dan kejernihan pada uji plaque assay. Kesimpulan: Bakteriofag yang diisolasi dari limbah IPAL menunjukkan aktivitas litik terhadap Staphylococcus aureus secara in vitro. Hasil ini memberikan dasar ilmiah awal bagi pengembangan penelitian lanjutan mengenai pemanfaatan bakteriofag sebagai agen biologis potensial dalam pengendalian Staphylococcus aureus pada infeksi sendi periprostetik.

Introduction: Periprosthetic joint infection (PJI) is a serious complication following hip and knee arthroplasty, with Staphylococcus aureus being one of the most common causative pathogens. Treatment failure is frequently associated with biofilm formation and increasing antibiotic resistance. These challenges have encouraged the exploration of alternative biological approaches, including bacteriophage therapy. Methods: This study was an in vitro experimental study. Hospital wastewater samples from the wastewater treatment plant (WWTP) of Dr. Cipto Mangunkusumo National General Hospital were used as sources for bacteriophage isolation. The lytic activity of bacteriophages against Staphylococcus aureus was evaluated using the double agar overlay (plaque assay) method. Lysis zones (plaques) and variations in plaque morphology were observed as indicators of lytic activity. Results: Bacteriophages were successfully isolated from hospital wastewater samples and demonstrated lytic activity against Staphylococcus aureus. The presence of plaques with varying sizes and clarity indicated lytic interactions between bacteriophages and Staphylococcus aureus under in vitro conditions. Conclusion: Bacteriophage isolates obtained from hospital wastewater exhibited lytic activity against Staphylococcus aureus in vitro. These findings provide preliminary evidence supporting further research on bacteriophages as potential biological agents for controlling Staphylococcus aureus associated with periprosthetic joint infection.
