Analisis Kadar MDA pada Ginjal Tikus Nefropati Obstruktif yang Diberikan 6-Gingerol = Analysis of MDA Levels in the Kidneys of Obstructive Nephropathy Rats Treated with 6-Gingerol.
Latar Belakang: Nefropati obstruktif merupakan kelainan pada fungsi atau struktur ureter yang mengganggu aliran urin normal. Kondisi ini berhubungan dengan progresivitas penyakit ginjal kronis (PGK) karena dapat menyebabkan inflamasi dan fibrosis tubulointerstisial sehingga terjadi ketidakseimbangan antara antioksidan, ROS, dan peroksidasi lipid. Bahan aktif dari jahe, yaitu 6-gingerol yang memiliki manfaat antiinflamasi, antioksidan, dan antifibrosis diketahui memiliki potensi farmakologis untuk pengobatan nefropati obstruktif. Studi ini bertujuan untuk meneliti kadar malondialdehyde (MDA), suatu parameter peroksidasi lipid pada tikus nefropati obstruktif yang diberikan intervensi 6-gingerol. Metode: Model unilateral ureteral obstruction (UUO) dilakukan dengan meligasi ureter kiri pada tikus Sprague-Dawley jantan berusia 8 minggu dengan berat 120-150 gram. Tikus lalu diberikan intervensi 6-gingerol dosis 200 mg/kgBB/hari selama 14 hari secara peroral. Delapan belas ekor tikus dibagi menjadi kelompok sehat (NN), kontrol negatif (UUO), dan kelompok perlakuan (6G) yang diberikan 6-gingerol. Di akhir minggu ke-2, tikus diterminasi menggunakan ketamine-xylazine lalu jaringan ginjal kiri diambil. Selanjutnya dilakukan pemeriksaan MDA menggunakan metode Thiobarbituric Acid Reactive Substances (TBARS) lalu larutan diperiksa menggunakan spektrofotometer. Hasil: Hasil studi menunjukkan 6-gingerol dosis 200 mg/kgBB/hari dapat mengurangi kadar MDA secara bermakna (p < 0,05) pada ginjal tikus kelompok 6G dibanding kelompok UUO. Kesimpulan: Studi ini berhasil membuktikan bahwa 6-gingerol mampu menurunkan kadar MDA pada ginjal tikus model UUO.

Introduction: Obstructive nephropathy is an abnormality of ureter function and structural that disrupts normal urine flow. This condition is associated with the progression of chronic kidney disease (CKD) due to its ability to induce inflammation and tubulointerstitial fibrosis, thereby causing an imbalance between antioxidants, ROS, and lipid peroxidation. 6-Gingerol, an active compound derived from ginger, has antiinflammatory, antioxidant, and antifibrotic properties, suggesting its pharmacological potential for the treatment of obstructive nephropathy. This study aims to investigate the effects of 6-gingerol intervention on malondialdehyde (MDA) levels, a parameter of lipid peroxidation, in rats with induced obstructive nephropathy. Method: A unilateral ureteral obstruction (UUO) model was established by ligating the left ureter of 8-week-old male 120-150 gram Sprague-Dawley rats. The rats were administered 6-gingerol at a dose of 200 mg/kg BW/day orally for 14 days. A total of 18 rats were divided into three groups: a healthy control group (NN), a negative control group (UUO), and a treatment group (6G) with 6-gingerol intervention. At the end of the 2-week period, the rats were euthanized using ketamine-xylazine and left kidney were collected. MDA levels were measured using Thiobarbituric Acid Rective Substances (TBARS) and the solutions were analyzed using a spectrophotometer. Results: The study results demonstrated that 6-gingerol dosage of 200 mg/kg BW/day significantly reduced MDA levels (p < 0.05) in the kidney tissues of the 6G treatment group compared to the UUO group. Conclusion: This study successfully found that 6-gingerol effectively reduced MDA levels in the kidneys of rats with a UUO-induced model of obstructive nephropathy.
