Identifikasi DNA Mucorales pada jaringan biopsy sebagai upaya pengembangan diagnosis mukormikosis = Identification of Mucorales DNA in the tissue biopsy as part of diagnostic development of mucormycosis.

Mukormikosis adalah infeksi jamur yang disebabkan jamur ordo Mucorales. Diantara genus yang tergabung dalam ordo Mucorales, Rhizopus sp. merupakan genus yang paling sering menyebabkan infeksi (70%) dan diikuti genus Mucor serta Lichtheimia. Tujuan penelitian ini adalah mengembangkan diagnosis mukormikosis pada jaringan biopsi berbasis molekular. Sebanyak 17 sampel jaringan uji yang akan diisolasi untuk mendapatkan DNA jamur menggunakan KIT QIAGEN dengan menggunakan tiga pasang primer (ITS1-4, ITS1-2 dan ITS86-4). Hasil penelitian didapatkan bahwa identifikasi jamur Mucorales dari regio ITS dapat dilakukan pada sampel jaringan. Dari ketiga primer tersebut, ITS1- 4 hanya berhasil amplifiaksi (6/17) atau 35% sampel uji dan pada primer ITS1 -2 dan ITS86-4 dapat teramplifikasi dengan baik pada semua jaringan. Ketiga primer tersebut dapat digunakan dalam penegakan diagnosis mukormikosis berbasis molekular, dan primer terbaik dalam identifikasijamur Mucorales adalah ITS1- ITS2 dan ITS86-ITS4. Identifikasi berbasis molekular lebih baik dari pada metode konvensional (KOH dan kultur).

Mucormycosis is a fungal infection caused by fungi ofthe ordo Mucorales. Among thegenera belonging to the ordo Mucorales, Rhizopus sp. is the genus that most often causes infection (70%) and is followed by the genera Mucor and Lichtheimia. The aim of this study was to develop a diagnosis of mucormycosis in molecularbased biopsy tissue. A total of 17 test tissue samples will be isolated to obtain fungal DNA using the QIAGEN KIT using three pairs of primers (ITS1 -4, ITS1- 2, and ITS86-4). The results showed that the identification of Mucorales fungi from the ITS region could be carried out on tissue samples. Of the three primers, ITS1-4 only succeeded in amplification (6/17) or 35% ofthe test sample, and ITS1- 2and ITS86-4 primers were amplified well in all tissues. These three primers can be used in themolecular-based diagnosis of mucormycosis, and the best primers for the identification of Mucorales fungi are ITS1-ITS2 and ITS86-ITS4. Molecular-based identification is better than conventional methods (KOH and culture)
