Aktivitas Antiviral Ekstrak Etanol Daun Cassia alata terhadap Virus Dengue Serotipe-1 pada Tahap Pra dan Pasca Internalisasi Infeksi Sel Vero = Antiviral Activity of Cassia alata Leaf Ethanol Extract Against Dengue Serotype-1 at Pre and Post Entry Infection of Vero.
Dengue virus (DENV) is a virus that causes dengue fever and is endemic in tropical countries, including Indonesia. Currently, there are no DENV antivirals that have been approved by drug regulatory agencies. Therefore, it is important to look for DENV antiviral candidates, one of which comes from herbal ingredients. This study focuses on investigating the action of an ethanol extract of Cassia alata (CA) leaves in vitro on the inhibition of DENV serotype-1 replication. This experimental study used DENV-1 strain IDS 11/2010 (an Indonesian strain) and VeroE6 cells. IC50 and CC50 values were measured by focal assay (FFA) and 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyltetrazolium bromide MTT assays, respectively. Furthermore, an IFA assay was conducted to determine the decrease in the number of infected cells and an FFA assay to determine the decrease in virus production. Then, the inhibition of CA leaf extract at pre-infection and post-infection stages was identified. Based on the in vitro test, the IC50, CC50, and SI values of CA extract were 13.38 g/mL, 801.14 g/mL, and 59.89 g/mL, respectively. The ethanol extract of CA leaves at 2xIC50 concentration (26.75 g/mL) showed inhibitory ability at pre-entry and post- entry stages. Based on virus production analysis, the best inhibition was at the pre- post-entry stage using both IFA and FFA methods (84% and 78%, respectively). The ethanol extract of CA leaves is an herbal ingredient that can be developed as an antiviral drug for DENV in the future. Keyword: DENV-1, Antivirus, Cassia alata, in vitro, IC50, CC50.
