Pengaruh Pemberian Ekstrak Kedelai Kaya Lunasin pada Ekspresi Protein E-cadherin Jaringan Kolon Mencit yang Diinduksi AOM/DSS = Effect of Lunasin-Rich Soybean Extract on E-Cadherin Protein Expression on Colon Tissues of Mice Induced by Azoxymethane and Dextran Sodium Sulfate.

Latar belakang: Kanker kolorektal merupakan salah satu kanker dengan tingkat insidensi dan mortalitas tertinggi di dunia. Tingkat insidensi dan mortalitas tersebut terus mengalami peningkatan di negara berkembang. Hingga saat ini, masih belum ditemukan terapi yang efektif. Studi-studi zat antikanker menunjukkan potensi Lunasin yang terkandung dalam kedelai mampu mencegah perkembangan kanker. Pada penelitian ini, peneliti akan menguji pengaruh ekstrak kedelai kaya lunasin terhadap ekspresi ECadherin yang berperan dalam metastasis kanker kolorektal. Metode: 36 mencit Swiss Webster dibagi ke dalam enam kelompok. Lima kelompok diinduksi dengan azoxymethane (AOM) dan dextran sodium sulfate (DSS) selama dua minggu. Setelah itu, tiga dari lima kelompok tersebut diberikan ekstrak kedelai kaya lunasin dengan dosis 250 mg/KgBB, 300 mg/KgBB, dan 350mg/KgBB selama empat minggu. Mencit diterminasi pada minggu ke-6 dan dilakukan perwarnaan imunohistokimia E-Cadherin pada jaringan kolon distal mencit. Hasil: Terdapat perbedaan secara signifikan antara kelompok kontrol negatif dengan kelompok terapi lunasin dosis 300 mg/KgBB (p < 0,001) dan 350 mg/KgBB (p < 0,001). Kesimpulan: Pemberian ekstrak kedelai kaya lunasin dengan dosis 300 mg/KgBB dan 350 mg/KgBB dapat meningkatkan ekspresi E-Cadherin pada sel epitel kripta jaringan kolon mencit yang diinduksi AOM/DSS.

Introduction: Colorectal cancer is one of the cancers with the highest incidence and mortality rates in the world. The incidence and mortality rates continue to increase in developing countries. So far, no effective therapy has been found. Studies of anticancer substances show the potential of lunasin contained in soybeans to prevent the development of cancer. In this study, researchers will examine the effect of lunasin-rich soybean extract on the expression of E-Cadherin which plays a role in colorectal cancer metastasis. Method: 36 Swiss Webster mice were divided into six groups. Five groups were induced with azoxymethane (AOM) and dextran sodium sulfate (DSS) for two weeks. After that, three of the five groups were given so extract rich in Lunasin at a dose of 250 mg/KgBW, 300 mg/KgBW, and 350 mg/KgBW for four weeks. Mice were terminated at week 6 and E-Cadherin immunohistochemical staining was performed on the distal colon tissue of mice. Result: There was a significant difference between the negative group and the lunasin therapy group at a dose of 300 mg/KgBW (p < 0.001) and 350 mg/KgBW (p < 0.001) Conclusion: The administration of lunasin-rich soybean extract at doses of 300 mg/KgBW and 350 mg/KgBW can increase the expression of E-Cadherin in crypt epithelial cells of mouse colon tissue induced by AOM/DSS.
