Efek Lunasin pada Ekspresi NF-ĸB Kolon Mencit Diinduksi Dextran Sodium Sulfate dan Azoksimetana = Effect of Lunasin upon NF-ĸB Expression on Colon of Mice Induced by Dextran Sodium Sulfate and Azoxymethane.
Latar belakang: Kanker Kolon merupakan penyumbang terbesar keempat kematian akibat kanker global. Tatalaksana yang tersedia saat ini masih memiliki beragam efek samping. Lunasin diduga memiliki aktivitas terapeutik dengan menurunkan persinyalan NF-ĸB yang krusial dalam perkembangan kanker kolorektal. Diharapkan melalui penelitian ini, dapat diketahui efektivitas lunasin sebagai terapi kanker kolorektal. Metode: Bahan biologi tersimpan kolon mencit dibagi menjadi enam kelompok. Semua kelompok diinduksi DSS dan AOM kecuali kelompok normal. Pemberian aspirin dan dosis lunasin (250, 300, dan 350 mg/kgBB) dilakukan selama 5 minggu pada mencit. Mencit kemudian dikorbankan dan jaringan kolon diekstraksi. Selanjutnya pembuatan preparat sediaan dan pewarnaan imunohistomikia serta HE. Gambar spesimen diambil dengan Indomicroview dan penilaian H-score dengan Immunohistochesmistry profiler. Hasil: Semua kelompok uji berbeda signifikan secara statistik dengan kelompok negatif (p < 0,05). Kelompok kontrol positif (mean=133,66) berbeda signifikan dengan kelompok normal (mean=107,52), dosis 300 mg/kgBB (mean=179,73), dosis 250 mg/kgBB (mean=205,37), dan kelompok negatif (mean=247,34) namun tidak terhadap kelompok dosis 350 mg/kgBB (mean=141,06). Kesimpulan: Lunasin menginhibisi ekspresi NF-ĸB epitel kolon mencit yang diinduksi AOM dan DSS. Ditemukan bahwa peningkatan pemberian dosis lunasin berpengaruh pada peningkatan inhibisi ekspresi NF-ĸB. Pemberian dosis 350 mg/kgBB berpengaruh setara dengan aspirin (kontrol positif), sehingga dapat dijadikan dosis acuan untuk penelitian lanjutan lunasin sebagai terapi kanker kolon.

Introduction: Colon cancer is the fourth largest contributor to global cancer deaths. Currently available treatment still has a variety of side effects. Lunasin is known to have therapeutic activity by inhibiting NF-B signaling which is crucial for colorectal cancer progression. It is hoped that through this research, the effectiveness of lunasin as a colorectal cancer therapy can be known. Method: Stored biological material of colon of mice was divided into six groups. All groups were induced DSS and AOM except the normal group. Aspirin and lunasin doses (250, 300, and 350 mg/kgBW) were administered for 5 weeks to the mice. Mice were then sacrificed and the colonic tissue then extracted. Furthermore, the preparation of preparations and immunohistochemical staining and HE. Specimen images were taken with Indomicroview and H-score assessment with Immunohistochesmistry profiler. Result: All test groups were statistically significant with the negative group (p < 0.05). The positive control group (mean=133.66) was significantly different from the normal group (mean=107.52), 300 mg/kgBW dose group (mean=179.73), 250 mg/kgBW dose group (mean=205.37), and the negative group (mean=247.34) but not the 350 mg/kgBW dose group (mean=141.06). Conclusion: Lunasin inhibit NF-ĸB expression in AOM and DSS-induced mouse colonic epithelial. It was found that increasing the dose of lunasin had an effect on increasing the inhibition of NF-ĸB expression. Giving a dose of 350 mg/kgBW has the same effect as aspirin (positive control), so it can be used as a reference dose for further research on lunasin as colon cancer therapy.
