Peran elektroakupunktur di titik ST36 Zusanli terhadap stres oksidatif di jaringan hati tikus model intoksikasi alkohol akut = Role of electroacupuncture at ST36 Zusanli on oxidative stress in the liver tissue of acute alcohol intoxication mouse model.

Latar belakang: Intoksikasi alkohol akut (acute alcohol intoxication/AAI) merupakan kondisi berbahaya secara klinis akibat dari satu episode konsumsi alkohol yang sangat banyak melampaui metabolisme hati. Stres oksidatif memainkan peran penting dalam patogenesis cedera hati akibat alkohol. MDA dan GSH telah banyak digunakan sebagai biomarker stres oksidatif. Elektroakupunktur (EA) merupakan modifikasi dari akupunktur manual yang terbukti dapat menekan stres oksidatif. Tujuan: Untuk mengetahui apakah EA frekuensi rendah di titik ST36 Zusanli dapat menekan stres oksidatif di hati pada tikus model intoksikasi alkohol akut. Metode: Studi ini adalah studi eksperimental acak, tersamar tunggal, post test dengan kontrol, subjek dan pemeriksa laboratorium disamarkan. Dua putuh empat tikus Wistar jantan dibagi secara acak menjadi 4 kelompok masing-masing terdiri dari 6 tikus: (1) kelompok normal; (2) kelompok alkohol; (3) kelompok EA dan (4) kelompok sham. Penelitian dilakukan sejak Maret-Juli 2021 di Laboratorium Hewan Coba, Puslitbangkes Biomedis dan Teknologi Dasar Kesehatan, Badan Litbangkes, Kemenkes RI. Tikus diberikan alkohol dosis tunggal 6 g/kgBB intragastrik untuk membuat model AAI. EA dan sham dilakukan satu kali selama 30 menit setelah tikus diberikan alkohol dan kehilangan righting reflex. Enam jam setelah pemberian alkohol, kadar MDA dan GSH hati diukur. Hasil: Rerata kadar MDA hati kelompok alkohol dan sham lebih tinggi secara bermakna dibandingkan dengan kelompok normal dan EA (p < 0,005). Tidak ada perbedaan bermakna antara rerata kadar MDA hati kelompok EA dan kelompok normal (p=1,000). Rerata kadar GSH hati kelompok EA dan kelompok normal lebih tinggi dibandingkan kelompok alkohol dan sham. Tidak ada perbedaan bermakna rerata kadar GSH hati kelompok EA dan kelompok nomal (p=0,993). Kesimpulan: EA frekuensi 2 Hz di titik ST36 Zusanli dapat menghambat stres oksidatif di hati pada tikus model intoksikasi alkohol akut.

Background: Acute alcohol intoxication (AAI) is a clinically harmful condition as a result of a single episode of excessive drinking over liver metabolism. Oxidative stress plays an important role in the pathogenesis of alcohol-induced liver injury. MDA and GSH has been widely used as a oxidative stress biomarker. Electroacupuncture (EA) is a modification of manual acupuncture which has been shown to suppress oxidative stress. Objective: This study was conducted to investigate the role of low frequency electroacupuncture (EA) at ST36 Zusanli acupoint on supress oxidative stress in rat liver tissue of acute alcohol intoxication model. Methods: This study was a randomized, controled, single-blinded, experimental posttest, with both subjects and laboratory investigators blinded. Twenty-four male Wistar rats were divided randomly into 4 groups of 6 rats each: (1) a normal group; (2) an alcohol group; (3) an EA group; and (4) a sham group. The study was conducted from March to July 2021 at Laboratorium Hewan Coba, Puslitbangkes Biomedis dan Teknologi Dasar Kesehatan, Badan Litbangkes, Kemenkes RI. Rats were given a single dose of alcohol 6 g/kgBW intragastrically to create an AAI model. EA and sham was applied once for 30 minutes after the rats were given alcohol and lost their righting reflex. Six hours after alcohol administration, liver MDA and GSH levels were measured. Results: The mean liver MDA levels in the alcohol and sham groups were significantly higher than the normal and EA groups (p < 0.005). There was no significant difference between the mean liver MDA levels in the EA group and the normal group (p=1,000). The mean liver GSH levels in the EA group and the normal group were higher than the alcohol and sham groups. There was no significant difference in the mean liver GSH levels in the EA group and the normal group (p=0.993). Conclusion: EA 2 Hz frequency at ST36 Zusanli acupoint attenuated alcohol-induced oxidative stress in the rat liver
