Pengaruh Pencucian Sperma dengan Metode Density Gradient Centrifugation Terhadap Aktivitas Spesifik Dinein ATPase Pada LakiLaki Infertil Astenozoospermia = The Effect of Sperm Washing with Density Gradient Centrifugation Method to Specific Activity of Dynein ATPase in Infertile Male with Asthenozoospermia.

LATAR BELAKANG: Inseminasi intrauterin atau IIU adalah salah satu tata laksana dari infertilitas. Pencucian spermatozoa dengan metode density gradient centrifugation atau DGC dilakukan untuk memisahkan spermatozoa yang dapat digunakan untuk IIU. Pencucian spermatozoa dapat berpengaruh pada aktivitas dinein ATPase, yang berperan dalam motilitas, namun nilai pasti dari aktivitas dinein ATPase pada spermatozoa astenozoospermia sebelum dan setelah pencucian DGC belum diketahui METODE: Pada sampel yang telah diperoleh dari 6 partisipan laki-laki astenozoospermia, dilakukan analisis semen dengan merujuk pada guideline WHO tahun 2010, preparasi fraksi aksonem, penentuan kadar protein, serta pengujian aktivitas spesifik dinein ATPase yang dilakukan sebelum dan setelah dilakukannya pencucian spermatozoa dengan metode DGC. Aktivitas spesifik dinein ATPase diukur dengan melihat kemampuannya melepaskan fosfat dari ATP. Penentuan kadar protein dilakukan dengan menggunakan spektrofotometer nanodrop, dan dilakukan uji statistik berupa uji t berpasangan atau uji Wilcoxon untuk melihat derajat kemaknaan, dengan nilai p < 0,05 menandakan hasil yang bermakna. HASIL: Ditemukan adanya peningkatan rerata dari konsentrasi, morfologi, kelajuan, dan persentase spermatozoa motil progresif pada kelompok astenozoospermia setelah DGC dibandingkan dengan sebelum DGC. Aktivitas spesifik dinein ATPase juga mengalami peningkatan yang bermakna pada kelompok setelah DGC dibandingkan dengan kelompok sebelum DGC. KESIMPULAN: Setelah pencucian spermatozoa dengan menggunakan metode DGC, terjadi peningkatan aktivitas spesifik dinein ATPase dan peningkatan persentase spermatozoa motil progresif

BACKGROUND: Intrauterine insemination, also known as IIU, is one of the treatments used for infertility. Sperm washing procedure is used to separate motile spermatozoa that are going to be used for intrauterine insemination. One of the methods used in sperm washing is the density gradient centrifugation or DGC, which could affect the activity of dynein ATPase, which is important for sperm motility. Therefore, this study is conducted to find out the effect of sperm washing with DGC method to the specific activity of dynein ATPase. METHODS: Samples obtained from 6 male participants with asthenozoospermia were analyzed according to WHO’s 2010 guideline, followed by axoneme fraction preparation, protein level measurement, and dynein ATPase specific activity test were performed before and after sperm washing with DGC method. Dynein ATPase specific activity is measured by its ability to free phosphate from ATP. Sample protein level is measured by using the nanodrop spectrophotometer. Paired t-test or Wilcoxon signed-rank test were performed to see the degree of significant, with p-value < 0,05 indicates a statistically significant result. RESULT: There was an increase of concentration, morphology, velocity, and motility of spermatozoa between asthenozoospermia group before and after sperm washing with DGC method. There is also a significant increase in the specific activity of dynein ATPase before and after sperm washing with DGC method. CONCLUSIONS: After sperm washing procedure with DGC method, there is a significant increase in dynein ATPase specific activity and the percentage of motile progressive spermatozoa.
