The Effect of Gallic Acid towards the Viability of Cultured Endometriosis Cells = Efek Asam Galat terhadap Viabilitas Sel Endometriosis.
Introduction. There is an urgency to find a treatment for endometriosis, since currently there is no cure for endometriosis that guarantees no recurrence. Since gallic acid and its synthetic derivatives have been known to reduce cell viability in several cancer cells, this study intended to investigate whether these substances also inhibit the viability of endometriosis cells.
Methods. Endometriosis cells were isolated and cultured from patients who had undergone laparoscopy surgery. After cells were 70-80% confluent, they were harvested and divided into 10,000 cells per well. Cells were given treatment of gallic acid, heptyl gallate and octyl gallate, each with three dosages of 25.6 μg/ml, 51.2 μg/ml and 102.4 μg/ml. All interventions were made in a set of triplo on four samples. MTS assay was used to observe the viability of the endometriosis cells.
Results. After the data was rationalised with control, all three substances showed inhibition of endometriosis cells viability. On all three substances, there was a significant inhibition at dosage 102.4 µg/ml. The highest inhibition was found at heptyl gallate dosage 102.4 µg/ml with 70.52% of endometriosis cell viability inhibition.
Conclusion. It is evident from the present study that gallic acid, heptyl gallate and octyl gallate, acted as a potential cell viability inhibitory agent towards endometriosis cells. The highest inhibitory effect was found at dose 102.4 µg/ml for all substances, with heptyl gallate imposing the most effect.

Pendahuluan. Terdapat sebuah urgensi untuk mencari pengobatan bagi endometriosis, karena saat ini belum ada pengobatan yang menjamin tidak terjadinya rekurensi. Berdasarkan kemampuan asam galat dan derivatnya dalam menurunkan viabilitas sel kanker, penelitian ini bertujuan untuk meninvestigasi kemampuan zat-zat tersebut dalam menginhibisi viabilitas sel endometriosis.
Metode. Sel endometriosis diisolasi dan dikultur dari pasien yang telah menjalani operasi laparoskopi. Setelah sel 70-80% konfluen, sel dipanen dan dibagi kedalam well yang masing-masing berisi 10.000 sel. Sel diberikan asam galat, heptil galat dan oktil galat,,masing-masing dengan tiga dosis yaitu 25.6 μg/ml, 51.2 μg/ml and 102.4 μg/ml. Seluruh intervensi dilakukan secara triplo pada empat sampel. MTS assay digunakan untuk menguji viabilitas sel endometriosis.
Hasil. Setelah data dirasionalisasi dengan kontrol, ketiga zat menunjukkan inhibisi terhadap viabilitas sel endometriosis. Pada ketiga zat, terdapat inhibisi yang signifikan pada dosis 102.4 μg/ml. Sedangkan inhibisi tertinggi ditemukan pada heptil galat pada dosis 102.4 μg/ml dengan persentase inhibisi sebesar 70.52%.
Kesimpulan. Penelitian ini menunjukkan bahwa asam galat, heptil galat dan oktil galat, berpotensi sebagai zat inhibitori terhadap viabilitas sel endometriosis. Efek inhibitori tertinggi ditemukan pada dosis 102.4 μg/ml untuk semua zat, dengan heptil galat sebagai zat dengan efek terbesar.
