Human Umbilical Cord-Derived Mesenchymal Stem Cells’ Proliferative Capacity and Viability in Phosphate Buffered Saline (PBS) Solution = Kapasitas Proliferasi dan Viabilitas Human Umbilical CordDerived Mesenchymal Stem Cells di Dalam Larutan Phosphate Buffered Saline (PBS).
Stem cell therapy has been implemented in Cipto Mangunkusumo General Hospital (RSCM), Jakarta. However, technical problems necessitate one to store the already prepared stem cells in a solution which best preserves them, without hampering their viability and functions (PDT), until the operation is ready. Thus, this experiment explored the effect of phosphate buffered saline (PBS), as a storage medium, on human umbilical cord-derived mesenchymal stem cells’ (hUC-MSCs) viability and PDT over 168 hours. The experiment used passage 9 hUC-MSCs that are placed into 8 groups – 0, 3, 6, 24, 48, 72, 96 and 168 hours – with 4 observations each. After the time of interest elapsed, a sample of the cells were taken to determine their viability and its average value, while the others were seeded at 10,000 cells in a multi-well plate. When the seeded cells reached 70-80% confluence, they were harvested and their PDTs, as well as the average value, were checked using a formula. All the obtained data failed to meet the assumption of normal distribution and/or homogenous spread when Shapiro-Wilk’s test and Levene’s test were used respectively.
