The Effect of Suspension in Physiologic Saline on the Viability and Proliferative Capacity of Whole Human Umbilical Cord Derived Stem Cells = Pengaruh Penyimpanan dalam Larutan Fisiologis Terhadap Viabilitas dan Kapasitas Proliferasi Human Umbilical Cord-Derived Mesenchymal Stem Cells.

Introduction: Stem cells have recently been a topic of interest due to their ability to self renew and versatility, alleviating diseases ranging from graft versus host disorder to liver cirrhosis. Unfortunately, administrative and technical issues encountered in clinical settings may delay the administration of stem cells. This calls for a temporary mode of storage for stem cells that will not only preserve the stem cells, but also allow them to retain their quality. This study, evaluates physiologic saline as a solution to suspend passage 9 human umbilical cord-derived mesenchymal stem cells (hUC-MSCs).
Methods: The parameters assessed in this study will be viability and proliferative capacity (measured in PDT) of hUC-MSCs in suspension over 168 hours, with 4 readings taken at each 0, 3, 6, 24, 48, 72, 96, and 168 hours. Statistical analysis of collected data is done to compare data to baseline.
Results: The MSCs suspended in saline retained viability >70% for up to 96 hours. Statistical analysis yielded significant difference with baseline from 24 hours onwards for viability and 6 hours for PDT. This means that the MSCs have started losing proliferative capacity from 6 hours.
Conclusion: In conclusion, hUC-MSCs can only be stored for 70% hingga jam ke-96. Analisis statistik menunjukkan perbedaan yang signifikan dengan baseline, yaitu mulai dari jam ke-24 untuk viabilitas serta jam ke-6 untuk PDT. Hal ini menunjukkan bahwa MSC mulai kehilangan kapasitas proliferasinya pada jam ke-6.
