Fraksi Air Daun Cosmos caudatus Kunth sebagai Antiviral Dengue Serotipe-2 secara In Vitro = In-vitro test of water fraction of Cosmos caudatus Kunth leaf as an antiviral for dengue serotype-2.

Virus Dengue masih menjadi salah satu permasalahan kesehatan dunia hingga saat ini. Hal ini dapat dilihat dari tingginya angka kejadian infeksi DENV, yaitu mencapai mencapai 390 juta kasus di seluruh dunia pada tahun 2010. Di Indonesia sendiri, kasusnya mencapai lebih dari 110.000 kasus pada tahun 2013. Belum ditemukannya obat anti-virus bagi penderita infeksi virus Dengue, menyebabkan tatalaksana penyakit ini hanya sebatas terapi suportif. Sehingga diperlukan penemuan obat anti-virus yang terbukti ampuh untuk mengobati pasien infeksi virus Dengue. Tujuan utama penelitian ini adalah mengetahui potensi antiviral dari fraksi air daun Cosmos caudatus Kunth terhadap DENV serotipe-2 strain New Guinea C pada sel Huh7-it1. Aktivitas antiviral dinyatakan dengan nilai indeks selektivitas yang didapatkan melalui perbandingan half-cytotoxic concentration (CC 50 ) dan half-inhibitory concentration (IC 50 ). CC adalah ukuran seberapa toksik suatu ekstrak terhadap sel Huh7-it1 yang nilainya didapatkan melalui metode MTT assay. Selain itu, nilai IC 50 50 yang ditentukan melalui focus assay adalah suatu ukuran yang menunjukkan kemampuan suatu ekstrak menghambat replikasi DENV2. Fraksi air daun C. caudatus memiliki nilai CC sebesar 156.3 µg/ml µg/ml, IC sebesar 9.3 µg/ml dan indeks selektivitas sebesar 16.6. Fraksi air daun C. caudatus memiliki potensi antiviral terhadap DENV2 sel Huh7it-1 dengan tingkat sedang. Diperlukan penelitian lebih jauh guna mengetahui senyawa aktif yang terkandung dalam fraksi air dan bagaimana mekanisme kerjanya terhadap DENV2. 50

Dengue virus (DENV) infection is still a massive health worldwide problem. This can be seen from the high number of DENV infection cases worldwide that reached 390 million worldwide. In Indonesia, the number exceeded 110.000 cases in 2013. The abscense of anti-viral drug to treat DENV2 infection, give rise to supportive treatment as the only treatment management. The discovery of a proven anti-viral drug is still needed to treat patients with Dengue virus infection. The main aim of this research is to identify the antiviral activity of water fraction of Cosmos caudatus Kunth leaves toward DENV serotype-2 New Guinea C strain at huh7-it1 cells. The substance antiviral activity is stated as selectivity index, which is the ratio of extract’s half-cytotoxic concentration (CC50 ) and extract’s half-inhibitory concentration (IC50 ). CC50 show how toxic an extract toward Huh7it-1 cells in which its value are determined by MTT assays method. Meanwhile, IC50 value show an extract’s capability to inhibit the replication of DENV-2. Water fraction of Cosmos caudatus leaves has the CC50 value of 156.3 µg/ml and IC value of 9.3 µg/ml hence its selectivity index was 16.6. Water fraction of C. caudatus leaves has a moderate level of antiviral activity against DENV-2 in Huh7it-1 cells. Further research is needed to unveil the active ingredient contained within the fraction and its mechanism of action as DENV-2 antiviral substance.
