Pengaruh Ekstrak Kedelai Terhadap Kadar MDA, GSH, Insulin, Konsentrasi Telomerase, Ekspresi Telomerase Revers Transcriptase (TERT) dan Jumlah sel  Pankreas pada Tikus Diabetes Mellitus yang diinduksi Aloksan. = The effect of Soybean Extract to Levels of MDA, GSH, Insulin, Telomerase Concentration, Telomerase Revers Transcriptase (TERT) Expression and the number of pancreatic -cells in the alloxan induced diabetes mellitus rats.

Latar belakang: Penelitian mengenai manfaat kedelai dalam penyembuhan penyakit diabetes mellitus sudah banyak dilakukan, namun belum diketahui pengaruh ekstrak kedelai terhadap peran protein TERT sel  pankreas. Tujuan penelitian: Mengukur kemampuan ekstrak kedelai dalam meningkatkan ekspresi TERT sel  pankreas pada tikus diabetes melitus. Metode Penelitian: Penelitian dilakukan di Animal House Departemen Farmakologi FKUI, pada bulan Januari 2013 sampai dengan Oktober 2013. Eksperimental dengan Randomized block design. Subyek : 60 tikus putih jantan galur Sprague-Dawley dikelompokkan secara acak menjadi 6 kelompok: tikus normal, Tikus DM (diinduksi aloksan), tikus DM + glibenkl (glibenklamid), tikus DM+ekstrak kedelai 1 mg/kgBB/hari, tikus DM+ekstrak kedelai 5 mg/kgBB/hari, tikus DM+ekstrak kedelai 25 mg/kgBB/hari. Analisis data dilakukan dengan menggunakan SPSS 20. Variabel yang diukur yaitu glukosa darah puasa (GDP), kadar MDA, GSH, aktivitas enzim telomerase, kadar hormon insulin, ekspresi TERT dan jumlah sel  pankreas. Hasil: GDP pada perlakuan dengan ekstrak kedelai menurun secara bermakna (p < 0,05) dibandingkan dengan tikus DM. Kadar MDA, kadar GSH, kadar hormon insulin dan konsentrasi enzim telomerase tidak berbeda dibanding tikus DM. Ekspresi TERT pada DM + 25 mg/kgBB/hari lebih tinggi secara bermakna p < 0,05 dibanding tikus DM, jumlah sel  pankreas pada tikus perlakuan ekstrak kedelai lebih tinggi secara bermakna p < 0,05 dibanding tikus DM. Kesimpulan: Ekstrak kedelai 1, 5 dan 25 mg/kgBB/hari menurunkan GDP dan meningkatkan ekspresi TERT sel  pankreas pada tikus diabetes mellitus yang diinduksi aloksan.

ABSTRACT Background: Studies about the benefits of soybean in the treatment of diabetes mellitus had been performed previously, but the effects of soybean extract on the role TERT protein  cells of the pancreas is not known. Objective: To investigate the effect of soybean extract in TERT expression of pancreatic  cells in diabetic rats. Method: The study was conducted at the department of Pharmacology FMUI, in January 2013 to October 2013. Sixty male Sprague-Dawley rats were randomly divided into 6 groups: namely normal rat, alloxan induced diabetes mellitus (DM) rat, DM rat + glibenclamide, DM rat + soybean extract 1 mg/kgBW/day, DM rat + soybean extract 5 mg/kgBW/day, DM rat + soybean extract 25 mg/kgBW/day. Statistical analysis was performed using SPSS 20. The measured variables were fasting blood glucose (FBG), MDA, GSH, activity of the enzyme telomerase, levels of the hormone insulin, expression of TERT and the number of pancreatic  cells. Result: FBG after treatment with soybean extracts decreased significantly compared with DM rats (p < 0.05). There was no differences in MDA, GSH levels, insulin hormone levels and telomerase enzyme concentration between soybean extract rats and control. TERT expression in DM rat + 25 mg/kgBB/day was significantly higher than DM rats (p < 0.05), and the number of pancreatic  cells in rat treated soybean extract was significantly higher than DM rats (p < 0.05). Conclusion: Soybean extracts decrease FBG and increase the expression of TERT  cells of the pancreas in alloxan induced DM rats.
