Pengaruh Ekstrak Daun Mesua ferrea Terhadap Titer Virus Dengue In Vitro = Effects of Mesua ferrea leaves Extract on Dengue Virus Titer In Vitro.

Infeksi virus dengue (DENV) saat ini menjadi masalah infeksi yang paling besar di seluruh dunia dalam 50 tahun terakhir. Masalah ini terjadi di seluruh dunia dan 70% dari seluruh kasus terletak di Asia Tenggara, ditambah lagi angka fatality case di Indonesia akibat DENV termasuk yang tertinggi. Meski demikian hingga saat ini belum ada antivirus yang efektif yang mampu melawan infeksi DENV. Ekstrak daun Mesua ferrea merupakan ekstrak tanaman yang diduga memiliki efek antiviral terhadap DENV. Dalam penelitian ini akan dicari daya hambat antiviral ekstrak daun Mesua ferrea (IC 50 ) serta efek sitotoksiknya terhadap sel Huh7it-1 (CC 50 ), dan angka indeks selektivitas (SI). Dalam penelitian ini digunakan sel Huh7it-1 yang diinfeksi DENV dan dalam penelitian ini dipakai konsentrasi ekstrak sebesar 320, 160, 80, 40, 20, dan 10 g/mL. Nilai SI didapatkan dengan cara membagi nilai CC 50 . Pada proses analisis data digunakan uji Kruskal-Wallis dengan uji post hoc MannWhitney. Dari hasil penelitian didapatkan angka IC 50 dari ekstrak daun Mesua ferrea adalah 118,49 g/mL, angka CC 50 sebesar 189,68 g/mL, dan angka SI sebesar 1,6. Jika dibandingkan dengan hasil penelitian sebelumnya angka SI sebesar 1,6 dirasa relatif kecil dan dapat disimpulkan bahwa ekstrak daun Mesua ferrea tidak terlalu berpotensi sebagai antivirus DENV.

Dengue viral infection today is still a huge burden in the world over the last five decade. This problem has affected the world and 70% of all cases happened in South East Asia. Furthermore Indonesia has higher fatality case compared to other countries. Nevertheless the antiviral for DENV has not been found yet. Mesua ferrea leaves extract is one of the natural extracts that may has antiviral effect to DENV. In this research we figured out inhibitory concentration of Mesua ferrea leaves extract to DENV and the toxicity to Huh7it-1 cells. This research tried to find selectivity index (SI) of Mesua ferrea leaves extract. Huh7it-1 cells were used as host cells. The concentration of the extract that we used in this research was 640, 320, 160, 80, 40, 20 and 10 g/mL. Using Kruskall-Wallis with Mann-Whitney as a post hoc for the data analysis, we found that the IC 50 of Mesua ferrea leaves extract 118,49 g/mL, CC 50 value189,68 g/mL, and SI was 1,6. Compared to prior research, the SI of Mesua ferrea leaves extract relatively low, therefore it’s not too potential to be used as the antiviral for DENV.
