Kemampuan sari mengkudu (Morinda Citrifolia) dalam menangkal stress oksidatif akibat pemberian CCI4 pada tikus = The effect of Morinda citrifolia extract in minimizing the oxidative stress on rats induced with CCI4.
Field and methodology: Morinda citrifolia, known as mengkudu in Indonesia, has long been known to have therapeutic properties like anti bacterial, anti viral, anti fungi, anti cancer, analgetic, anti inflammatory, and imunomodulatoiy. Nowadays, it is also being said to have antioxidant property. Based on that opinion, this experiment is conducted, in which 30 male rats (Wistar strain), weighed approximately 200 grams each, were divided as group I (control), group II (CCI4) and group III (morinda extract + CCI4) with 10 rats in each group. For the period of 14 days standard diets were given to those three groups, with group II received single dose of CCI4 (55 mg/gram body weight) on the 12th day, while group III, in addition to standard diets, also received morinda extract 2 ml per day for 12 days. Four hours after the administration of morinda extract on the 12th day, rats in group III also received CC14 with the same dose as group II. On the 14th day, all rats in those three groups were sacrificed for the measurement of malondialdehyde (MDA) in blood and liver and glutathione in liver and erythrocytes. Histopathology of the liver examination by haematoxylin-eosin staining was also performed in this study. Results and conclusions: The liver and blood MDA concentrations in group III (56,16 ± 3,13 nmol/g and 12.02 ± 0.63 nmol/ml respectively) were significantly lower than those in group II (85,19 ± 4,35 nmol/g and 17.00 ± 0.97 nmol/ml). Liver and erythrocyte glutathione in group III (1,62 ± 0,07 umol/g and 107,56 ± 8,44 nmol/gHb respectively) were significantly higher than those in group II (0,96 ± 0,04 nmol/g and 69,92 ± 3,43 nmol/gHb). On the other hand in histopathologic examinations, the degrees of liver cell injuries in group III (x = 0,8) were statistically ower than those in group II (x = 2,8), but showed no significant difference compared to those in group I (x= 0,6). From these results, it could be concluded that the administration of Morinda citrifolia extract to rats could prevent, or at least minimize the toxic effect of CCI4.
Key words: Morinda citrifolia, MDA, glutathione, liver histopathology.
