Ekspresi Gen Viral Infectivity Factor (Vif) HIV-1 pada Escherichia Coli BL21. = Expression of Viral Infectivity Factor (vif) Gene of HIV- 1 in Escherichia Coli BL21.

Gen viral infectivity factor (vif) memiliki peran esensial dalam patogenesis HIV ke sel manusia melalui interaksinya dengan protein APOBEC3G. Oleh karena itu, karakterisasi gen dan protein vif menjadi fondasi penting dalam rangka penemuan obat atau vaksin HIV yang baru berbasis vif. Ekspresi merupakan proses produksi protein dari gen. Pada penelitian ini, plasmid rekombinan pGEX-6p-1 yang telah disisipi gen vif HIV-1 diperoleh dari peneliti sebelumnya. Escherichia coli BL21 sebagai vektor ekspresi dan IPTG sebagai inducernya. Pada penelitian ini, digunakan berbagai macam kondisi seperti konsentrasi akhir IPTG maupun lama inkubasi, untuk menghasilkan protein VIF. Hasil ekspresi dibaca dengan menggunakan metode elektroforesis pada gel poliakrilamid. Pada hasil, belum didapatkan pita protein VIF. Faktor-faktor yang mempengaruhi tidak terbentuknya protein VIF turut dibahas dalam penelitian ini.

Viral infectivity factor (vif) gene has an essential role in the pathogenesis of HIV into human cells through its interaction with APOBEC3G protein. Therefore, the characterization of vif genes and vifproteins become an important foundation in order to find a drug or a new HIV vaccine based on vif pathway. Expression is the process of protein production from genes. In this study, recombinant plasmid pGEX-6p-1 which has been inserted by HIV-1 vif gene is obtained from previous researchers. Escherichia coli BL21 is used as an expression vector and IPTG is used as the inducer. In this study, variety of conditions such as IPTG final concentration and incubation period is used to produce VIF protein. Expression results are read using the method of polyacrylamide gel electrophoresis. In the results, VIF protein bands have not been obtained yet. Factors that influence the absence of VIF protein in this research are discussed.
