Protein expression of nucleoprotein (NP) gene of H5N1 avian influenza A virus
Avian influenza is caused by influenza A virus subtype H5N1. According to Depkes RI, from 2005 to 2009 cumulatively there have been 161 cases in Indonesia, with 134 fatalities. This research has the aim of producing the NP (Nucleoprotein) protein that is part of the virus, since virus proteins or antigens that can be produced from recombinant DNA will be needed in the development of diagnostic kits. This is done using E. coli bacteria strains containing the plasmids with the genes coding for the NP protein. The bacteria are induced with 0.3 mM IPTG to express the proteins, and electrophoresis is performed to analyze the protein size. Comparing the proteins bands from the cultures containing the NP protein gene and those lacking the gene, a new thick band of protein around the size of 54 kDa is found in the samples with the NP gene induced for two hours or more, and is suspected to be the band containing NP protein.
