Efek ekstrak air akar acalypha indica linn. dosis 15 mg pada kelumpuhan sambungan neuromuskular bufo melanostictus schneider akibat penyuntikan pankuronium bromida 0,1 mg

Dengan meningkatnya penderita penyakit autoimun saat ini, banyak peneliti menaruh perhatian terhadap penyakit tersebut. Salah satu penyakit autoimun adalah Myasthenia Gravis. Obat- obatan yang digunakan untuk pengobatan penyakit ini belum ada bukti bahwa obat tersebut benar- benar aman dan yang terjangkau. Di Indonesia banyak tanaman obat yang mempunyai efek terhadap berbagai penyakit, namun belum ada bukti tentang tanaman yang berefek sebagai neuroterapi. Tanaman tersebut adalah akar kucing (Acalypha indica Linn.). Penelitian ini bertujuan untuk membuktikan efek neuroterapi ekstrak air akar Acalypha indica Linn. secara in vivo pada katak Bufo melanostictus Schneider, pada 3 kelompok percobaan, yaitu: kontrol negatif berupa pemberian air, kontrol positif diberi piracetam, dan kelompok ekstrak dengan dosis 15 mg. Setiap katak awalnya disuntik Pankuronium bromida 0,1 mg sebagai pelumpuh otot dan ditunggu sampai katak menjadi kolaps. Setelah itu katak diberi perlakuan sesuai kelompoknya. Efek neuroterapi ditentukan dari kemampuan katak untuk pulih kembali setelah mengalami kelumpuhan. Dari hasil analisis dengan menggunakan uji one way anova ditemukan terdapat perbedaan bermakna antara kelompok kontrol negatif (air) dan kelompok ekstrak 15 mg (p = 0,004), dan terdapat perbedaan tidak bermakna antara kelompok kontrol positif (pirasetam) dengan kelompok ekstrak 15 mg (p = 0,257). Melalui penelitian ini disimpulkan bahwa ekstrak air akar tanaman Acalypha indica Linn. dengan dosis 15 mg memiliki efek neuroterapi secara in vivo yang sebanding dengan pirasetam.

With the increase of patients with autoimmune disease, researcher have put a lot of concern on these diseases. Myasthenia Gravis is one of the autoimmune disease. The drugs that were used for the treatment of Myasthenia Gravis have no evidence to be really safe and had an affordable price. Indonesia has a lot of herbal medicines that have effect on some disease, but not yet proven to have neurotherapeutic effect . Akar kucing (Acalypha indica Linn.) is one of them. This research is aimed to prove neurotherapeutic effect of Acalypha indica Linn’s root extract in vivo on Bufo melanostictus Schneider, with three experimental groups: negative control group which use water, positive control group using piracetam and 15 mg dose of extract group. Each frog is injected with Pancuronium bromide 0,1 mg as a muscle relaxant first and wait for the frog to collapse. After that, it will get a treatment depends on its group. The neurotherapeutic effect was determined by the ability of the frog to recover after it collapse. With one way anova test, a significant difference is found between the negative control (water) and the 15 mg extract group (p = 0,004), and an unsignificant difference is found between the positive control (piracetam) and the 15 mg extract group (p = 0,257). From this research, it is concluded that Acalypha indica Linn’s 15 mg root extract has a neurotherapeutic effect in vivo that is equal to piracetam.
